- Abstract
- 본 발명은 리파제를 코딩하는 핵산을 포함하는 리파제 분비 발현 카세트로서, 상기 리파제는 서열번호 1의 아미노산 서열에서 N-말단으로부터 52 번째 및 190 번째 아미노산 중 어느 하나 이상이 다른 아미노산으로 치환된 리파제 분비 발현 카세트; 이를 포함하는 발현 벡터; 이를 포함하는 형질전환 미생물; 상기 형질 전환 미생물을 배양하는 단계 및 리파제를 회수하는 단계를 포함하는 리파제의 제조방법; 및 전이에스터 활성 및 분비 발현능이 증가된 변이형 리파제에 관한 것이다. 본 발명의 분비 발현 카세트, 발현 벡터 및 이를 포함하는 형질전환 미생물을 이용하여, 전이에스터 활성이 개선된 리파제를 고효율로 생산할 수 있으며, 생 산된 리파제는 전이에스터 반응이 필요한 모든 공정에 유용하게 이용될 수 있다.
