dc.description.abstractWe report a new highly specific and sensitive interleukin-4(IL-4) bioassay by the measurement of CD23(FcfRff) expression on B-cells using fluorescence activated cell scanner(FACScan). This assay is based on the quantitative analysis of membrane CD 23 on B-cells by double staining with monoclonal anti CD23(Leu 20:FceRH ) antibody-phycoerythrin conjugate and monoclonal anti CD20(Leu 16 : pan B marker) antibody-FITC conjugate. This method overcomes the problems associated with the conventional B-cell proliferation bioassay by providing a significantly higher specificity for IL-4. The sensitivity of this assay is also proven to be greater than IL-4 ELISA (en-zyme-linked immunosorbent assay) or B-cell proliferation bioassay, especially when heterogenous test samples such as human serum or T-cell culture supernatant were used. Development of this system as an IL-4 bioassay will be useful for the analysis of IL-4 in the patients with IL-4 related diseases such as allergy, hypersensitivity and nephrosis.-
