- Abstract
- Monoclonal antibodies against phospholipids, especially bioactive phospholipids, give very important information to understand cellular signal transduction and receptor conformation. A series of monoclonal antibodies that recognize phosphatidylcholine, phosphatidylserine and phosphati-dylinositol-^l-monophosphate were established by intrasplenic immunization method. Originally, this technique v,as the method of choice for the immunization with very little amounts (about nanogram) of immunogen. However, using this method, I immunized phospholipids which have been known to have poor immunogenicity (hapten) due to their low molecular weight, and attained successful outcomes. Moreover, 8-mercaptoguanosine administration showed a considerable increase in hybri-doma production and PC(phosphatidylcholine) specific monoclonal antibodies. In future, this technique may be helpful to produce monoclonal antibodies which have been difficult to make until now.
