dc.description.abstractIn Saccharomyces cerevisiae^ expression of an allantoin degradative gene, DAL7, is induced by allophanate. The regulation of DAL7 gent; expression involves interactions between exacting elements including upstream activation sequence (UAS), upstream induction sequence (UIS) and upstream repression sequence (URS), and transacting factors such as DAL81, DAL82 and DAL80 proteins. The binding properties of DAL80 to the d卜acting elements in the DAL7 were investigated by gel retardation ami DNase I footprinting experiments using the DAL80 protein which was fiised to C-terminal/regon of glutatirione S-transferase (GST). The DAL80 protein fiised to GST was demonstrated to bind to UAS and URS2 sequences, which suggested that the DAL80 protein may activate transcription of DAL7 both by acting as a GATAA UAS binding factor and by reKeving repression. Also, the DAL80 protein seemed to be associated with a 52-kDa protein through protein-protein interaction.-
