In process of drug developments, many fields of studies should be combined and coupled. Chemistry, pharmacy, pharmacology,toxicology and clinicah; are the necessary fields for new drug development. In the view of pharmacologist and toxicologist, drug discovery and development strategy is divided into two categories. First category is a in vitro and in vivo screening and second is a preclinical and clinical evaluation of new drug candidates. Really, many compounds could be found by screening steps, but nearly all-of them was discarded by downstream works such as toxicity testing and clinical trials. Accordingly, to save time and fund required for drug development, the establishment and development of meaningful screening method are essential works. Animal cells and human cells are widely used for in vitro evaluation of drugs and toxicants. In present paper, the developments and applications of human hepatocytes, lymphocytes will be presented and discussed. For primary culture of hepatocyte, animals induding rat, mouse, dogs were used for cell sources. In the case of animals, whde Ever could be obtained and collagenase perfusion used to isolate hepatocytes. Owing to the limitatic的 of sam^e supply, human hepatic parenchymal cells were isolated from surgical biopsy samptes of liver by nonperfusion method. Hepatocytes were cultured successfully in mcxiolayer for longer than 12 days. Isolated and cultured hepatocytes were examined on their morphological and functional characteristics 班id proved to maintain their integrity at a reasonable level in all testing items. In vitro regeneration of cultured hepatocyte was analyzed and their modulating factors were examined chemically and physically. The growth phenomena of human hepatocyte were introduced to the evaluation of hepatotoxicity of some toxic agents and drugs. The toxicities induced in hepatocytes by TCDD, CC1O ethanol were compared with normal human fibroblast and transformed hepatoma cell line.
