The monokines produced by activated monocyte/macrophage and reactive oxygen species (ROS) and lysozyme produced by neutrophil play an important role in acute and chronic inflammation The role of neutrophil in the inflammation needs to be reexamined because cytokines are found to modify the neutrophil function. To investigate which monokine is responsible for the neutrophil activation, neutrophils were preincubated with recombinant human 1L-1ß, TNF-? or 1L-6 for 30min and stimulated subsequently with PMA for 10 min. These preincubation with monokines mark-edly enhanced PMA induced ROS production and lysozyme release(priming effect). In the LPS-treated monocyte/macrophage culture supernatant, activities of 1L-1, TNF and 1L-6 were found by bioassay. ROS production and lysozyme release were markedly enhanced by preincubating neutrophils with LPS-treated culture supernatant, which might be due to synergistic actions of monokines produced in the supernatant.
