dc.description.abstract | Beta amyloid (Aβ) is a potent oral immunogen with potential utility as a vaccine against Alzheimer’s disease (AD). Our previous work investigated the ability of a protein comprised of five tandem repeats of Aβ42(5Aβ 42) to induce an immune response in a transgenic AD mouse model, and demonstrated that immunization with potato-derived 5Aβ42 reduced the Aβburden as well as many of the AD-associated pathological features. To provide a potato-derived 5Aβas vaccine material, the stable yield of 5Aβprotein through constant expression of introduced transgene is the most important factor. In the present study, we confirmed that the 5Aβ42 transgene were inherited and stably expressed in in vitro vegetative propagation over a period of 3 years without silencing. The 5Aβ42 protein, too, was expressed well without change of its antigenicity in transgenic potato. Stable storage conditions were vital in maintaining the stability of the potato-derived 5Aβ42 protein, and activity of the protein was not affected following up to three months’ storage at low temperature. | - |
